Stealth Rock would get it down by about 30% HP since it's Super Effective, and then Toxic would take care of the rest.
But yeah, conventional means of attacking it would just be impossible as long as it has Rest, even only using Super Effectives.

it is slow so one hit with e.q. then really another so most likely u will be dead

Unless EQ is STABed with a Pokemon of 400+ Attack, it'd probably take three EQs to seal the deal, 500 Defense is very impressive. Hell, my Forretress needs three EQs to kill it, and I think it caps at 367 Defense. Granted, it has far higher HP than Shuckle.
A Rest Shuckle that Defense buffs itself would be a pain in the ass though, you're just better off with letting Toxic do all the work for you, it only has like 264 HP after all.

Poisen jab. Thats what i'd use, coz it can poisen you and it's a good attacking move.

Well, not really.
Poison is .5 damage to Shuckle, and shuckles usually have leftovers/rest. Leftovers will neutralize the poison, and rest will recover attack based damage. O.o
Water Rock and steel would be the only hopes. Not poison.
